“Onco-Innovations is a Canadian preclinical-stage precision oncology company. It’s headquartered in Calgary, and we’re developing a first-in-class DNA Damage Response inhibitor against a novel target,” Mohamed said. “That’s a DNA repair enzyme called polynucleotide kinase phosphatase, or PNKP. As far as we’re aware, there is no clinical-stage PNKP program anywhere in the world, and we hold exclusive worldwide rights to the University of Alberta’s foundational PNKP inhibitor patents together with an exclusive sublicense to the nanoparticle delivery platform that accompanies it.”

Mohamed then explained how ONC010 is designed to address a longstanding challenge with radiosensitizers and chemosensitizers by combining biological and physical mechanisms intended to selectively target cancer cells and tumor tissue.

“I would say, in one word, the big difference is selectivity,” he said. “ONC010 stacks two independent selectivity mechanisms. The first is biological selectivity. Synthetic lethality limits the drug’s lethality to cells that have already lost PTEN or SHP-1. It limits the lethality to only those cells, and healthy cells that have not lost them don’t experience this toxic effect of the drug… The second is a physical selectivity. The nanoparticle concentrates the drug within tumor tissue rather than distributing it uniformly in the body because the tumor vasculature is permeable or leaky in a way that normal vasculature isn't.”

Turning to the preclinical evidence supporting ONC010, Mohamed highlighted a University of Alberta study involving the nano-formulated PNKP inhibitor that the company intends to advance into human testing.

“In that experiment, the encapsulated drug demonstrated single-agent antitumor activity, it was tolerated at the doses tested, and it prolonged survival relative to controls,” Mohamed said. “This is a really critical outcome, and it was one of the reasons why I wanted to proceed with supporting this further as a drug candidate. The drug worked on its own. The treated animals lived longer than the controls did, and the drug was tolerable at the maximum feasible dose… That one experiment validates the whole drug development thesis all at once.”
